Description
BGC20-0761 is a novel, orally active tryptamine analogue with potent 5-HT6 and 5-HT2A receptor antagonist activity, conferring antipsychotic and cognition-improving properties.

Schizophrenia is a chronic, severe and disabling psychiatric disorder that is estimated to affect ~1% of the world’s population. It is treated with atypical and typical antipsychotic pharmaceuticals. These are generally efficacious against the positive symptoms such as delusion and hallucination, but are not usually effective against the negative symptoms such as depression, irritability and loss of concentration. The unmet medical need is for new drugs that alleviate both positive and negative symptoms and have fewer side-effects.

The pharmacological profile of BGC20-0761, as determined in a number of preclinical studies, suggests that it should address the positive symptoms and also the negative and cognition aspects of schizophrenia, with the potential for a low level of side-effects. Preliminary pharmacokinetic studies also suggest the potential for once daily dosing with a high CNS penetration.
